Just curious if Organic Potting Soil could be used as a substrate (even temporarily) for a small Eastern box turtle? Thanks in advance!

Possibly, but read the packet to find out what is actually in it and check for additives. No fertilisers, no sand, no charcoal, no white bits whatever they’re for.
Organic does not mean it is plain top soil, peat or coco coir.
Top soil tends to compact hard and doesn’t make a great substrate as it compacts hard when damp.
