should the wood of my Russian tortoises enclosure be real or fake wood, also how often should I change the substrate completely

You can use either, assuming fake means chipboard, plywood or hardboard.

You need to line it with something water resistant like a pond liner or shower curtain either way.

Depends what substrate you use.
I use coco coir and it lasts practically forever, just needs spot cleaning, really.
