Pick up any (fresh!)poop found in the enclosure. A warm bath in a container large enough for your tortoise to walk around in could also help you get a sample.

Put the sample in a closed container so it doesn't dry out. Samples from several days would be perfect, as not everything is expelled every day. With several poop samples you increase the chance of a correct diagnosis.
